I’ve used MediaWiki and Instiki in the past, and didn’t really like either of them.
What’s the best out there today?

I currently use 3: wetpaint, wikispaces, and pb wiki. I got used to wikispaces and can’t change in the middle of the year, but I prefer wetpaint.
i’m partial to PBWiki. It’s really easy to use and they have a bunch of “plug ins” that allow you to easily add interactive widgets like video, audio and productivity tools to your wiki page. Here’s more info.
BTW, PBWiki is looking for alpha testers for the new version of PBWiki, you can get more info here.
I’ve used wikispaces with relative success—haven’t tried any others. Here’s a couple examples of studies we’ve done:

We also have, in progress, separate studies on Hamlet and Macbeth.
I’ve found that using wikis provides motivation for the “digital natives” I’m teaching to work in their own ‘language,’ encourages them to collaborate (especially those that are more reticent in classroom discussions) and promotes deliberative discussion in which the conversation is well thought out. We maintain the spontaneity of classroom discussion in parallel.
